Biological Description

Specificity Anti-Calcium Pump PMCA4 ATPase Mouse Antibody [E6J21] detects endogenous levels of total PMCA4 ATPase protein. This antibody is specific to the PMCA4 isoform.
Background Plasma membrane calcium ATPases (PMCAs) are essential regulators of calcium-dependent signaling pathways, functioning through interactions with partner proteins. By exporting calcium ions out of the cell, PMCAs maintain low local concentrations of intracellular free calcium, thereby enabling precise control of signaling events within specific cellular subdomains. These pumps are critical for sustaining proper cytoplasmic calcium homeostasis through continuous calcium extrusion into the extracellular space. The PMCA family consists of four isoforms (PMCA1–4), each encoded by a distinct gene. While PMCA1 and PMCA4 are expressed broadly across tissues, PMCA2 and PMCA3 display more restricted expression patterns limited to specialized cells. Beyond calcium clearance, PMCAs achieve functional specificity by engaging their intracellular domains in protein–protein interactions with diverse cytoplasmic partners. Among them, plasma membrane calcium ATPase 4 (PMCA4) has emerged as a key modulator of cancer biology, influencing processes such as cell migration, proliferation, and resistance to apoptosis. Notably, PMCA4 is overexpressed in pancreatic ductal adenocarcinoma (PDAC), where its elevated levels are associated with poor clinical outcomes.
