Biological Activity

Description Imperatorin (Ammidin, Marmelosin, Pentosalen, 8-Isoamylenoxypsoralen), a naturally occurring psoralen-type furanocoumarin, is a potent myorelaxant agent and acts as a calcium antagonist on vascular smooth muscle.
In vitro
In vitro Imperatorin has estrogen-like activities in MCF-7 cells and stimulatory effects on ALP activity of Saos-2 osteoblast cells[1].

Method

The MCF-7 cells are seeded into six-well plates in standard growth medium at a density of 100 000 cells per well and allowed to attach overnight (24 h). The growth medium is replaced with phenol red free DMEM containing 10% charcoal-dextran stripped human serum (hormone-free) for 72 h, after which the medium is refreshed and chemicals are added. After a further 24-h exposure, the cells are harvested and washed with PBS. Cell pellets, which are obtained by centrifugation at 1000 rpm for 15 minutes, are stained with 1 mL of freshly propidium iodide (PI) staining solution (0.1% of Triton X-100, 0.1% of sodium citrate, 50 μg/mL of PI and 10 μg/mL of RNase A in PBS). Flow cytometric analysis of cell cycle distribution is performed.
In Vivo
In vivo Imperatorin acutely administered at the doses of 10 and 20 mg/kg impairs anxiogenic effect of nicotine in the elevated plus maze (EPM) test. Imperatorin acts as irreversible inactivator of GABA-transaminase and increases the GABA content in the brain. Co-administration of subthreshold doses of imperatorin and nicotine produces an improvement inmemory processes in animals[2].
